VISUALISATION OF RESPIRATORY ACTIVITY IN BREATHING SLICES FROM MICE USING VOLTAGE-SENSITIVE DYES

The investigation of the spatial sequencing of neuronal activity in the respiratory network during normal and impaired activity requires high temporal resolution. Voltage sensitive dyes meet these requirements and can be used to identify the exact timing of activity in different elements and regions of the respiratory network.

We have been able to established voltage imaging in the rhythmic slice preparation from the pre-Bötzinger complex of mice. In this slice preparation we have used di-4-ANEPPS. Recordings were performed at an unprecedented speed of 100Hz and above using spike triggered averaging with a CMOS-camera (MiCAM Ultima, Japan). The speed of the image acquisition was fast enough to show that the activity in the pre-Bötzinger complex is preceding the activity in the hypoglossal nucleus.

We conclude that voltage imaging in slice preparation is a valuable tool to analyze changes of spatial pattern in different mouse models of respiratory disturbances.
