THE II-III LOOP OF CAV2.3 E-TYPE VOLTAGE-GATED CA2+ CHANNEL INTERACTS WITH HSP70 WHEN OVER-EXPRESSED IN HEK-293 CELLS

Voltage-activated Ca^2+ channels participate in Ca^2+ dependent processes such as synaptic differentiation, secretion, transmitter release, and neuronal plasticity. Some signalling cascade start at cytosolic domains of the ion conducting subunit (Ca\ alpha1) which interacts with target proteins directly, in a Ca^2+ dependent or independent manner. The cytosolic loop between domains II & III of Ca_ v2.3 confers a novel Ca^2+ sensitivity to E-type Ca^2+ channels.To understand Ca^2+ and phorbol ester mediated activation of Ca_v2.3, proteins interacting with II-III loop were identified. The II-III loop of human Ca_ v 2.3 was over-expressed in HEK293 cells, and hsp70, which is known to interact with PKC, was identified as a novel interaction partner. Immunopurified II-III loop-proteins of neuronal and endocrine Ca_ v2.3, stimulate autophosphorylation of exogenous PKCa, suggesting that hsp70 may act as an adaptor protein for the Ca^2+ dependent targeting of PKC to E-type Ca^2+ channels.
