ABUNDANT EXPRESSION OF THE TRANSCRIPTION FACTOR LHX6 IN FAST-SPIKING GABAERGIC INTERNEURONS IN MOUSE DENTATE GYRUS

GABAergic interneurons are diverse and can be divided in various types based on their morphological, physiological properties and the expression of Ca2+-binding proteins (e.g. parvalbumin). Most cortical interneurons are derived from progenitors in the ganglionic eminence which migrate tangentially to their final destination, where they differentiate into diverse types. However, little is known about the mechanisms generating this diversity. Accumulating evidences suggest that interneuron types are generated by the action of distinct transcription factors. One of them, the homeobox gene Lhx6, was recently shown to be specifically expressed in the medial ganglionic eminence which gives rise to neocortical fast-spiking parvalbumin-positive interneurons. To test the hypothesis that Lhx6 is expressed in distinct interneuron types, we used Lhx6- EGFP transgenic mice. We performed whole-cell patch-clamp recordings of EGFP-labelled dentate gyrus interneurons in acute brain slices to determine their morphological and physiological properties. Our data show that 70% of the recorded cells (n=48) fired action potentials at high frequency (130±10 Hz at 850 pA, 32UC) and showed mainly axon arborizations in the granule cell layer. Thus, our data indicate that Lhx6-EGFP is abundantly expressed in hippocampal fast-spiking basket cells.
