Anti-inflammatory effect of thrombolytic preparation longolytin

Inflammation is the defence reaction, connected to different proteolytic systems of organism (kallikrein-kinin, hemostasis, fibrinolysis, etc.). Proteases as known are able to stimulate inflammation and on other hand to reduce it. There are variety protease's prepatations with anti-inflammatory action. Longolytin is proteases complex of fungi origin with high thrombolytic activity, studied in different animal models. This work is about its anti-inflammatory effect.
Methods: Inflammation process was caused by procedure of thrombus formation in rabbit's year marginal vein. The venous segment (10–12 mm) was restricted with two special clamps (serphines) for thrombin administration, and thrombi was forming for 1–1.5 h. Strong prolonged constriction injured tissues around thrombotic vessel, caused inflammation following redness, edema, disappearance of native capillaries drawing. Thrombotic vein and inflammation area was applicated with longolytin and its mixture every day for 7 days and changes were observed by visual control in four groups of rabbits:with longolytin only; with control glycerol; with mixture longolytin and heparin; witn heparin only.
Results: The inflammatory changes correlated with process of thrombolysis. In one group longolytin application caused in 5–6 times acceleration of thrombolysis time, compared to control glycerol group. Simultaneously finishing of inflammatory reaction decreased from 7–15 days in control to 4–12 days. The mixture longolytin and heparin had maximal thrombolytic and anti-inflammatory effect (2–10 days)compared to heparin action (5–14 days).There were dissapeared edema, redness and capillaries drowing became normal 3 times faster, than in glycerol application, twice faster than in heparin group. Longolytin (proteases complex) has thrombolytic and anti-inflammatory action.
