The high platelet activity plays a key role in arterial thrombosis. ADP is one of the most important mediators of platelet aggregation in vivo. Effect of ADP on platelet activity is mediated by receptors P2Y1 and P2Y12. Co-activation of the P2Y1 and P2Y12 is necessary for activation of GPIIb-IIIa receptor and complete platelet aggregation. In our study the P2Y1, P2Y12, GPIIb and GPIIIa gene expressions and their effect on platelet activity were investigated. We formed a group of 65 volunteers including 31 healthy donors and 34 patients with ST-elevation myocardial infarction, the age from 21 to 80 years, 44 male and 21 female and analyzed the P2Y1, P2Y12, GPIIb and GPIIIa gene expression as level of mRNA by RT-PCR using the TaqMan assay in ABI Prism 7000 Sequence detection System, cDNA prepared from total RNA isolated from platelets, ADP-induced platelet aggregation by optical method (2.5, 5 and 10mkM ADP) as level of aggregation (T,%) and rate of aggregation (V,%/min). We found that the mRNA levels of the investigated genes were widely varied – from 0.005 to 19.53 for P2Y1, from 0.07 to 23.72 for P2Y12, from 0.02 to 15.54 for GPIIb and from 0.004 to 9.65 for GPIIIa. The positive Spearman correlation between P2Y1 and GPIIIa gene expressions and between P2Y12 and GPIIb gene expressions were revealed – R = 0.9 (P = 0.006) and R = 0.34 (P = 0.06), respectively. Except this levels of the GPIIIa and P2Y1 gene expression have a positive Spearman correlation with T(%) and V(%/min) of ADP-induced aggregation (10mkM) – R = 0.58 (P = 0.08) and R = 0.71 (P = 0.047), respectively. In contrast, the P2Y12 gene expression had inverted Spearman correlation with V(%/min) of ADP-induced aggregation (10 mkM) – R = −0.44 (P = 0.05). In conclusion, the expressions of the platelet receptor genes may be associated with each other and influenced the platelet activity.
