Abstract
Introduction: Circulating platelets are heterogeneous with respect to their size, density and reactivity. Therefore, it is important to study of platelet volume in cord blood for understanding the platelet heterogeneity.
Methods: We examined peripheral blood cell counts, platelet indices, colony asays and CD34 positive cells in 85 cord bloods. An experienced obsterician did collecion of CB at delivery into the bags contained 28 ml CPD-A. We also examined aged controls (AC, mean 73 years) and younger volunters (YC, mean 38 years) for blood cell counts and platelet indices to compare the differences among the three groups. Anticoagulatnts were used EDTA for AC and 3.2% citrate for YC.
Results: In cord blood, mean platelet count (PLT), MPV, platelet-large cell ratio (P-LCR), platelet distribution width (PDW), erythrocyte count (RBC) and leukocyte count (WBC) were 33.1x10e4/ul, 9.3 fl, 19.1%, 9.9 fl, 447x10e4/ul, 14,660/ul, respectively. PLT of CB was significantly increased than that of AC, and MPV was significantly larger than that of YC. PLT had significant negative correlations with platelet indices in CB and AC (p<0.02). Platelet indices had positive correlations with RBC and significantly with CB volume (p<0.02). Significant positive correlation between PLT and WBC was observed in AC(p<0.0001), but not in CB. In AC, platelet indices had positive correlations with WBC. In CB, PLT and platelet indices had no correlations with WBC. Significant negative correlation between RBC and RET. Significant positive correlations between WBC and RBC were observed in AC, but not in CB. Total CFU-GM and CD34 positive cells had negative with PLT and positive correlations with platelet indices, but not significantly so.
Conclusions: Platelet volume of CB is larger than that of adult peripheral blood. Platelet indices will be influenced by erythropoiesis. WBC did not influence platelet indices in CB. There will be the different mechanisms in the platelet heterogeneity between CB and peripheral blood of adults.
