Abstract
Introduction: This paper describes inhibitory effect of DJW in PAF-induced aggregation as biological modifier.
Methods: DJW was extracted with 70% MeOH and the resulting I was dissolved with DW and II was obtained. Remaining fraction was extracted with EtOAc and resulting III, IV were obtained. ATP and PGE2 was detected by a bioluminescence method and by ELISA. The dose-response curves were deduced from fitting of values according to the logic function employing the term y=a+b/(l-(x/c)d), where x represents DJW concentration, y the pharmacologic effect expressed as % of control, and a, b, c and d are fitting constants. The IC50 values were calculated from individual fitted dose-response curves.
Results: The effects observed with total DJW was synergistic over-additive rather that additive since the sum of single contributions was lower than the effect of the total extract. III was specially protected against the lethality of PAF. Exogenously applied AA led to a 89% platelet aggregation, the release of 14 pmol of ATP, and the formation of either 225 pg of TXA2 or 45 pg of PGE2, each parameter being related to 106 platelets. An application of DJW 5 min before AA, dose-dependently diminished aggregation, ATP-re lease, and the synthesis of TXA2 and PGE2, with IC50 values of 70, 87, 65 and 72 g/ml, respectively. The similarity of the IC50 values suggests the inhibition of COX by DJW as the primary target.
Conclusions: DJW shows anti-thrombotic action on platelets and inhibits the action of PAF in vivo by an antagonistic effect on PAF.
