Performance of a new molecular assay for detection and differentiation of Bordetella spp. DNA

Objective: To investigate the performance of the new molecular artusTM Bordetella LC PCR Kit (QIAGEN Hamburg GmbH, Germany) assay that provides detection and differentiation of Bordetella spp. DNA in a routine diagnostic laboratory.
Methods: The assay was performed according to the manufacturer's package insert. A total of 85 nasopharyngeal swabs obtained from patients with a clinical presentation compatible to B. pertussis infection were tested with the new assay in an International Standards Organization (ISO9001, 2000)-certified routine clinical laboratory.
Results: Sixteen of 85 samples were found positive for Bordetella DNA. Differentiation by melting curve analysis yielded B. pertussis in 7 samples, B. parapertussis in 1 sample, and B. bronchiseptica in another one. In the remaining 7 samples, Bordetella DNA was detected by analysis of fluorescence curves of IS481 and IS1001 amplification products only. The heterologous internal control was consistently detected. In 7 specimens inhibition of the assay was found.
Conclusion: In conclusion, the new molecular assay includes all features required for molecular detection and differentiation of Bordetella spp. DNA. It proved to be suitable for the routine diagnostic laboratory allowing a rapid and safe diagnosis of pertussis.
