USSC EVOKES SIGNIFICANT CARDIAC REGENERATION AFTER MYOCARDIAL INFARCTION

Introduction: 

Stem cell-based therapy for ischemic cardiac disease so far has shown limited functional benefits and, particularly, structural repair has not well been documented. Here we explored whether there is cardiac regeneration after transplantation of unrestricted somatic stem cells (USSCs) into the rat heart after infarction.

Method: 

USSCs were isolated from human umbilical cord blood and injected intramyocardially into the border zone of the infarcted hearts after the LAD was occluded for 60-min. The structural and functional changes were analyzed histologically and echocardiographically 4-weeks after transplantation. Additionally, at day 7 the fate of USSC, replication of cardiomyocytes (CMs) with BrdU, role of epicardial stem cells and infiltration of immune cells were examined.

Results: 

USSCs significantly improved cardiac function by restoring ejection fraction to 62% (wall thickness: 2870 mm) as compared to 38% (wall thickness: 1290 mm) in untreated hearts (n= 8–10, p

Conclusion: 

USSCs evoked substantial structural and functional repair of the injured myocardium most likely by immune cell-mediated stimulation of epicardial stem cells.
