EARLY TREATMENT WITH A NATURAL ANTIOXIDANT POLYPHENOLIC COMPOUND IN EXPERIMENTAL TYPE-1 DIABETES: A NEW APPROACH FOR PREVENTING CARDIAC PARENCHIMAL AND PROGENITOR CELL DYSFUNCTION IN DIABETIC CARDIOMYOPATHY

The progressive damage in cardiac stem/progenitor cells (CSPC) plays a role in the pathophysiology of diabetic cardiomyopathy (DCM). We tested the hypothesis that Resveratrol (RSV) administration exerts a protective role on myocardial environment and CSPC survival and function.

Adult Wistar rats (n=128) with streptozotocin-induced diabetes were either untreated (D group; n=50) or injected with Resveratrol (2.5 mg/Kg/day, i.p.; DR group; n=58). Twenty rats were used as control group (C). Hemodynamics, cardiomyocyte mechanics and intracellular calcium dynamics were evaluated after 1, 3 or 8 weeks of hyperglycaemia. Myocardial remodelling and changes in tissue environment were also determined. The impact of diabetic "milieu" on CSPC turnover was analyzed in co-cultures of healthy CSPCs and myocytes isolated from DR and D hearts.

A marked loss of CSPCs and adult cardiac cells occurred during the first 3 weeks in D animals. Ventricular dysfunction appeared after 8 weeks and included a significant (p<0.01) decrease in: (i) ±dP/dt, (ii) myocyte fraction of shortening, (iii) rate of cell shortening and relengthening, and (iv) calcium transient amplitude. RSV administration prevented cell loss and preserved the functional properties of CSPCs and mature cardiomyocytes, reduced tissue inflammation and the unfavourable ventricular remodelling leading to a recovery of ventricular function.

In conclusion, RSV might constitute an adjuvant therapeutic option in DCM prevention and treatment.
