OXIDATION OF HUMAN RED BLOOD CELLS BY A FREE RADICAL INITIATOR: EFFECT ON RHEOLOGICAL PROPERTIES

Abstract: 

The main aim of the study was the investigation of the rheological properties of erythrocytes obtained from subjects admitted to the blood donors, quantifying lipid profile of HDL-C, LDL-C, Triglyceride and Total Cholesterol. The deformability index of erythrocytes was determined by using a dynamic stress rheometer (DSR-500). An increased rigidity of the erythrocytes in subject with higher lipid ratio TOT-C/HDL-C was found, compared with the erythrocytes in healthy subject at lower lipid ratio TOT-C/HDL-C. In order to explain the reason for the reduced deformability, the erythrocytes of subject with higher lipid ratio TOT-C/HDL-C were studied for the valuation of anionic permeability on membrane, hemolysis induced from AAPH (2,2-azo-bis (2-amidinopropane) dihydrochloride) hydrosoluble radical initiator, as well as the levels malonyldialdehyde. The different susceptibility to hemolysis by AAPH solution, indicated a decrease globular resistance, furthermore, the elevated value of malonyldialdehyde may suggest a lipid peroxidative damage in certain pool of the erythrocyte membrane in blood circulation.
